Hieff Unicon™ Robust Hotstart Taq DNA Polymerase, Glycerol-free (20 U/μL) is an antibody-mediated hot-start DNA polymerase with high amplification sensitivity and specificity. The blocking antibody is fully inactivated within 30 seconds at the initial denaturation temperature, releasing DNA polymerase activity. This hot-start mechanism helps suppress amplification caused by nonspecific primer annealing. The glycerol-free formulation is designed specifically for preparing lyophilized reagents. Features Lyophilization-Ready: Glycerol-free liquid formulation optimized for lyophilized reagent production. High Inhibitor Tolerance: Compatible with complex samples (swabs, blood); lyophilized beads support full-volume reconstitution with nucleic acid extracts. High Performance: Antibody-mediated hot-start ensures sensitive and specific amplification. Robust Stability: Retains >90% activity after 7 days at 37°C or 20 freeze-thaw cycles. Strong Stability Two batches of 14331 were evaluated for accelerated thermal stability and repeated freeze–thaw stability, with material stored at -20℃ as the control. After 7 days at 37℃ or 20 freeze–thaw cycles, both batches retained more than 90% of the activity measured under standard storage conditions. Figure 1. Accelerated thermal stability and freeze–thaw stability of two batches of 14331. 2. African Swine Fever Virus Detection with Lyophilized Beads Item Content Reagents 14331 was combined with qPCR buffer, ASFV primers/probes, and lyoprotectants to prepare 25 μL lyophilized qPCR reagents Samples Eight farm whole-blood samples were extracted by magnetic beads; 400 μL input, 70 μL elution Testing Lyophilized reactions were reconstituted with 5 μL or 25 μL nucleic acid extract, topped up to 25 μL with water Results 25 μL reconstitution gave Ct values ~2–4 cycles earlier than 5 μL, indicating tolerance to higher extract input and improved sensitivity Table 1. Detection of eight field samples using 14331-based lyophilized qPCR reagents. 3. HPV Detection with Lyophilized Beads Item Content Reagents 14331 was combined with qPCR buffer, HPV primers/probes, and lyoprotectants to prepare 15 μL lyophilized qPCR reagents Samples Ninety-eight HPV clinical samples were extracted by magnetic beads; 200 μL input, 100 μL elution Testing Each lyophilized reaction was fully reconstituted with 15 μL nucleic acid extract Results Both 14331-based and comparator reagents detected all 98 samples; 14331 gave lower Ct in 43, equal in 24, and higher in 31 Table 2. Detection of 98 HPV clinical samples using 14331-based lyophilized qPCR reagents.
