Benzamidine agarose resin is an affinity chromatography medium used for the purification of trypsin-like serine proteases. The resin consists of benzamidine—a synthetic competitive inhibitor of serine proteases—covalently coupled to a cross-linked agarose bead support, usually through a spacer arm. Benzamidine is a small, positively charged molecule that mimics the side chain of arginine/lysine and binds reversibly in the substrate-binding (S1) pocket of trypsin-like enzymes such as trypsin, thrombin, plasmin, urokinase, factor Xa, and related proteases. This allows these proteases to be selectively captured from complex mixtures while non-target proteins are washed away. BiCell Scientific’s benzamidine affinity resin uses 4% highly cross-linked agarose matrix with a mean size of 90 μm and capable of operating under 0.1 MPa (1 bar/14.5 psi). The hydrophilic nature of the matrix ensures low levels of non-specific binding leading to low levels of host cell-derived impurities in the elution pool. This resin binds 20-40 mg trypsin/mL resin and is suitable for use in batch mode, with gravity flow, or using a chromatography system.
