cFluor^® V547 Anti-Human CD42b (HIP1)

The mouse monoclonal antibody clone HIP1 reacts with human CD42b, also known as GPIbα (glycoprotein Ibα), a 135–145 kDa type I transmembrane glycoprotein and member of the leucine-rich repeat (LRR) family. CD42b forms a disulfide-linked heterodimer with CD42c (GPIbβ), which associates noncovalently with CD42a (GPIX) and CD42d (GPV) to constitute the GPIb-IX-V receptor complex on the platelet surface. The extracellular domain of CD42b contains eight leucine-rich repeats and an anionic region with sulfated tyrosines that collectively mediate binding to von Willebrand factor (VWF) exposed at sites of vascular injury, initiating platelet tethering and rolling under high shear conditions. CD42b also binds thrombin, coagulation factors XI and XII, high molecular weight kininogen, P-selectin, and integrin Mac-1 (CD11b/CD18). CD42b expression is restricted to platelets and megakaryocytes in bone marrow. Mutations and polymorphisms in CD42b are associated with Bernard-Soulier syndrome, a congenital bleeding disorder, and platelet-type von Willebrand disease. The HIP1 clone recognizes an epitope within the N-terminal region of the GPIbα chain and has been reported to inhibit ristocetin-dependent VWF binding and ristocetin-induced platelet agglutination. This antibody is reported for use in flow cytometric analysis, western blotting, immunohistochemical staining of frozen tissue sections, and in vitro functional blocking assays. For flow cytometric staining, the suggested use of this reagent is 5 µL per 1 million cells in a staining volume of 100 µL. If whole blood is analyzed, then use 5 µL per 100 µL. It is recommended that users titrate the antibody to obtain the optimal result for their specific application. Please briefly centrifuge the reagent vial before use.
