Cellecta's DriverMap™ Cell-Free Adaptive Immune Receptor (cfAIR) RNA & DNA TCR–BCR Profiling Kit (Human) is an easy-to-run, single-day, multiplex PCR-NGS assay that profiles the T-cell and B-cell receptor repertoire from human plasma. Compared with the bulk DriverMap™ AIR RNA and DNA kits, which start from blood cells or tissue, the cell-free AIR assay starts with cell-free nucleic acid samples from plasma to profile fragmented cell-free RNA (cfRNA) and cell-free DNA (cfDNA) from a shorter ~170 bp CDR3 region of all seven TCR and BCR chains (TRA, TRB, TRG, TRD, IGH, IGK, IGL) from at least 1 ml of plasma. Key Features Combined cfDNA + cfRNA immune receptor profiling: Two barcoded hybridization reactions give two signals from the same plasma sample. cfDNA-based TCR/BCR sequences capture lymphocyte turnover (apoptosis and necrosis), while cfRNA TCR/BCR sequences measure transcripts from extracellular vesicles (EVs) which reflect lymphocyte activation. Key biomarkers are obtained without invasive biopsies. Quantitative detection: Amplicons for each primer are tagged with Validator Barcodes (VBCs), which allows one to count template molecules and correct errors in the data deconvolution step. Fig 1 shows the number of TCR/BCR clonotypes detected in the plasma sample from healthy PBMC Control RNA and three types of cancer patients. One Kit, Multiple Sample Types: Matched control RNA from buffy coat/PBMC or FFPE tissue may also be profiled using the same protocol to identify tumor-derived and bystander clonotypes in the cfRNA or cfDNA repertoire. Each kit includes TCR and BCR primer sets, enzymes, buffers, a Positive Control RNA and custom NGS sequencing primers. Cell-free nucleic acid (cfNA), which combines cfDNA & cfRNA yields 10-fold more clonotypes than cfDNA alone. Matched FFPE tumor RNA yields a higher number of clonotypes due to tumor lymphocyte content, with overlapping clonotypes in FFPE and cfNA indicating apoptosis and immune activation.
