PEmax represents a significant architectural optimization of the PE2 system. By incorporating an additional Nuclear Localization Signal (NLS) and a refined nCas9 domain, it achieves superior nuclear import and targeting efficiency. Unlike traditional CRISPR methods, PEmax enables precise genomic modifications without inducing double-stranded breaks (DSBs), thereby maximizing editing yield and broadening the scope of targetable base conversions with enhanced fidelity.
